Evaluation of basic characteristics of thermal water with separation methods

The presence of inorganic ions is one of the criteria for thermal water assessing. The most important anions that affect the water quality and allow their use for therapeutic purposes, include sulfates, chlorides, nitrates and bicarbonates. Ions (as sodium, potassium, calcium and magnesium) are the significant cations. The ion chromatography is used for separation of these substances and also some methods based on electrophoretic migration of ions in an electric field. The electromigration method of isotachophoresis was chosen for determination of selected ions in water samples collected from thermal boreholes in Pasohlávky.
Pesticide glyphosate and possibilities of its determination

This bechelor thesis deals with determination of glyphosate employing capillary isotachophoresis technique. The literature review is devoted to pesticide, their distribution and use, properties and methods for the glyphosate determination, capillary zone isotachophoresis and its use. Experimental part focuses on the application of capillary isotachophoresis for determination of glyphosate, for determination of glyphosate in commercial preparations and testing of glyphosate sorption in soil.

This bachelor thesis deals with determination of organic acids in wine by capillary zone isotachophoresis method. The theoretical part is devoted to the properties of wine, its composition, its benefit to human health and capillary zone isotachophoresis and its practical use. The practical part deals with determination of organic acids in wine by capillary zone isotachophoresis. Organic acids are determined in three phases of white and red wine production.
Epitachophoretic preconcentration of proteins

In this work, we present a developed method for preconcentration of standards of proteins cytochrome c, myoglobin, and hemoglobin. Firstly, the electrolyte system was selected and optimized to provide low analysis time, narrow shape of the protein zone, and eliminate overheating at the center of the device. In the next step, the developed method was tested on the preconcentration of proteins: cytochrome c, myoglobin, and hemoglobin. After several adjustments myoglobin and hemoglobin were also uccessfully preconcentrated. After evaluation of pilot results, this technique provides up to fiftyfold preconcentration and recovery of over 70 % for each protein. The next plan is to preconcentrate proteins from complex biological matrices such as urine or blood plasma.
Isotachophoretic determination of ketone bodies and lactate in cattle serum

The aim of the work was the optimization of isotachophoretic determination of lactate and ketone bodies (beta-hydroxybutyrate and acetoacetate) for veterinary medicine. The measurement of these metabolites is very important for the evaluation of metabolic diseases of cattle. The complete separation of these three analytes together with other possible blood components was achieved using 10 mmol/l hydrochloric acid with beta-alanine as a leading electrolyte at pH = 3.15 and 10 mmol/l sodium propionate as a terminating electrolyte. Limits of detection determined from the standard mixture for acetoacetate, lactate and beta-hydroxybutyrate were 3.75 mu mol/l, 3.96 mu mol/l and 7.46 mu mol/l, respectively.

Using of capillary isotachophoresis and on-line capillary zone electrophoresis for the determination of microcomponents in samples with complex matrices

Beginning of capillary isotachophoresis is in seventieth years last century. Theory of the isotachophoretic separation process has been elaborated during twentieth years, the instrumentations were developed and considerable number of applications were publicated in articles. ITP-CZE enables in the first step to inject a relatively large sample volume, to separate sample components and to remove undesirable components. In the second step a simplified and defined part of the sample is analysed. Real samples of sweeteners, human serum and urine were analysed.
